ABSTRACT

Background Snakebite is considered a neglected tropical disease by the World Health Organization. In Brazil, about 70% of the envenomation cases are caused by Bothrops snakes. Its venom may provoke hemorrhage, pain, necrosis, hemolysis, renal or cardiac failure and even death in victims. Since commercial antivenom does not efficiently neutralize the local toxic effects of venoms, natural products have been tested in order to provide alternative or complementary treatment to serum therapy. Therefore, the present study aimed to evaluate the ability of the seaweed Plocamium brasiliense and its active derivatives to neutralize hemorrhagic, edematogenic, hemolytic, coagulant and proteolytic activities of B. jararaca venom. Methods Specimens of P. brasiliense were collected in Rio de Janeiro state, Brazil, dried and submitted to oil extraction using four solvents of increasing polarities, n-hexane (HEX), dichloromethane (DCM), ethyl acetate (ETA) and hydroalcoholic solution (HYD). The solvents were evaporated, yielding HEX, DCM, ETA and HYD extracts. Further, all extracts were dissolved in dimethylsulfoxide. In addition, two monoterpenes (8-bromo-3,4,7-trichloro-3,7-dimethyl-1E, 5E-octadiene and 1,8-dibromo-3,4,7-trichloro-3,7-dimethyl-1E, 5E-octadiene) and a cholesterol fraction were isolated from the extract of P. brasiliense prepared in hexane. Algal samples were incubated for 30 minutes with B. jararaca venom, and then tested for lethality; hemorrhagic, edematogenic, hemolytic, coagulant and proteolytic effects. Results Most of the algal extracts inhibited the toxic effects with different potencies. The DCM extract was the most effective, since it inhibited all types of toxic activity. On the other hand, the HYD extract failed to inhibit any effect. ABSTRACT

The crude extract of the marine brown alga Dictyota guineensis was analyzed by high-resolution gas chromatography-mass spectrometry (HRGC-MS). Five diterpenes were identified: dictyol E (the most abundant diterpene), dictyotadiol, dictyoxide, isopachydictyol A and pachydictyol A, all diterpenes from the chemical group I, i.e., mainly prenylated derivatives of known sesquiterpene skeletons that result from a first cyclization of geranyl-geraniol between positions 1 and 10. These diterpenes are known for their activity against bacteria, fungi and other activities. The results characterize D. guineensis as a species that yields exclusively diterpenes from group I, with low oxidation and low structural complexity. On Brazilian coasts, only D. mertensii provides exclusively prenylated guaiane diterpenes. Although D. guineensis presents alternate branches and fixing by rhizoidal branches, it is easily distinguishable from D. mertensii by the much narrower stem, short stature and flabelliform habit of the former species. On the other hand, both species have been characterized as producers of diterpenes of group I, in particular, prenylated guaiane. However, D. guineensis has a majority dictyol E in the lipophilic extract, while D. mertensii produces more complex prenylated guaianes, like dictyol H.

ABSTRACT

The effect of a Brazilian algae extract and also a mixture of two secodolastane diterpenes (linearol/isolinearol) that were isolated from the marine brown alga Canistrocarpus cervicornis were evaluated against biological activities of Lachesis muta snake venom. In vitro assays showed that the crude extract and the diterpenes were able to inhibit the clotting and proteolytic activity induced by L. muta crude venom, but not the hemolytic activity. However, only the diterpenes inhibited the hemolysis caused by a purified phospholipase A2 previously isolated from L. muta venom, denoted LM-PLA2-I. Interestingly, the crude algal extract and the diterpenes were able to protect mice from hemorrhage induced by L. muta venom. Thus, we may conclude that marine algae are rich and powerful sources of molecules that may be used against L. muta accidents in order to improve treatment of envenomation by this snake.

ABSTRACT

Dictyotaceae are a rich source of secondary metabolites, especially terpenes. These natural products have been studied for their bioactivity for human and for their ecological role in nature. The present work highlights de diversity of the Dictyotaceae terpenes, emphasizing their bioactivities and the biogeography of their sources. The sesquiterpenes are found in Padina, Dictyopteris and Taonia. Although Dictyopteris and Padina can be found in all oceans, Taonia has a more restricted distribution. Diterpenes of mixed origin have been reported in Lobophora, Taonia and Stypopodium. Stypopodium is a typically tropical and warm temperate genus. Diterpenes with the spatane and seco-spatanes skeletons are known from Spatoglossum, Stoechospermum and Rugulopteryx. Spatoglossum is distributed over all the tropical marine world. Stoechospermum is known to occur in the Indian Ocean and Rugulopteryx is distributed mainly in Asia and on the Pacific Coast of Australia. The genus Dictyota contains diterpenes as the major metabolite and are present in all oceans, reaching even the cold temperate regions of northern and southern hemispheres. The terpenoids from Dictyotaceae exhibit bioactivities such as inhibition of herbivores and antifungal, cytotoxic, antibiotic, anti-inflammatory, insecticidal and antiviral activities.

Os radicais livres e outros derivados ativos do oxigênio são inevitavelmente co-produzidos em algumas reações biológicas e exercem papel fisiológico importante. No entanto, essas espécies reativas têm sido descritas como fatores que participam diretamente dos mecanismos fisiopatológicos relacionados com a continuidade e as complicações de diversos estados patológicos como a aterosclerose, a diabetes, o câncer, a artrite reumatóide, entre outros. Dessa forma, a terapia antioxidante preveniria o desenvolvimento e a progressão dessas complicações. As algas marinhas representam uma importante fonte de substâncias antioxidantes naturais, uma vez que têm sistemas de defesas antioxidantes bem desenvolvidos. O presente trabalho é uma compilação das pesquisas realizadas sobre a atividade antioxidante de produtos naturais marinhos e extratos de algas marinhas bentônicas.


Free radicals and other reactive species of oxygen are co-produced in some biological reactions and they play important physiological role, and nevertheless they are reported as factors that take straight part in the pathophysiologic mechanism associated with continuity and complications of the several pathological process such as arteriosclerosis, diabetes mellitus, cancer, and arthritis, among others. In this way, the antioxidant therapy should prevent the development and progress of these complications. Seaweeds can be valuable source of natural antioxidant compounds since they have a well-developed antioxidant defense system. The present work is a compilation of the antioxidant activities of marine natural products and benthonic marine seaweeds crude extracts researches.
